في عام 635 ميلادية، فتحت الجيوش العربية الإسلامية بلاد الشام، وكانت هذه بداية المرحلة الذهبية لمدينة دمشق، فقد أصبحت عام 661 عاصمة للدولة الأموية التي امتدت من الأندلس في الغرب إلى بلاد الهند في الشرق، وأضحت أهم مركز للثقافة العربية والإسلامية واستمرت في لعب هذا الدور الحضاري حتى انتقلت عاصمة الدولة الإسلامية إلى بغداد عام 750 مع وصول العباسيين إلى الخلافة.
كانت دمشق هدفاً للعديد من محاولات الغزو الخارجي، أهمها حملات الصليبيين والمغول بين القرنيين الثاني عشر والخامس عشر. في عام 1516 احتل السلطان العثماني سليم الأول بلاد الشام، واستمر الاحتلال العثماني أربعة قرون تميزت بالتراجع الفكري والحضاري والعلمي.
